Drug Discovery Platform Based on Natural Products

 

Despite efforts to streamline syntheses of complex polycyclic natural products, many challenges remain, particularly for quaternary carbon center formation, making it difficult to advance these compounds to preclinical studies. This presentation will discuss synthetic strategies towards the total syntheses of quaternary spirocenter acorane sesquiterpenoids.

The acorane natural product, (+)-colletoic acid is a selective 11β-hydroxysteroid dehydrogenase type 1 inhibitor.

This important metabolic enzyme is responsible for converting cortisone to cortisol, which activates the glucocorticoid receptor (GR). The GR is essential for many cellular processes involve in metabolism and cancer.

Our synthetic efforts to this natural product along with our preliminary biological results will be presented.
